The effect of estrogen-containing combination oral contraceptives and chlormadinone acetate alone on plasma renin activity, renin substrate and aldosterone were compared in 16 women. Renin activity and substrate were increased on the estrogen combinations, most significantly with the highest dose of mestranol. Chlormadinone acetate had no such effect. Plasma aldosterone was, however, not significantly altered by any of the agents. The findings suggest the preferability of administering 17-acetoxy-progesterone derivatives to avoid hyperreninemia.
