Torsades de Pointes arrhythmias can be triggered by early after-depolarizations (EAD) . Mechanisms of generation of EAD's attributable to genetic mutations in the sodium channel, to changes in potassium currents, and to multiple changes in gene expression levels are reconstructed using computer models. One of these mechanisms has already been incorporated into whole organ models. Work is in progress to incorporate the other cellular mechanisms. Biophysically detailed models of this kind can identify combinations of receptor actions that would be expected to be therapeutic.
