Key result
Taking clopidogrel with food enhances bioavailability, increasing AUC ~9-fold versus the fasted state.
Why the study?
The effects of concomitant food intake on the bioavailability of a single oral dose of clopidogrel were not well characterized.
Does concomitant food intake affect the bioavailability of a single oral dose of clopidogrel in healthy male subjects?
Population
Healthy male subjects
Comparison
Single oral dose of clopidogrel fed vs fasted conditions
Design
Pharmacokinetic study with 14-day washout period

Food may markedly raise clopidogrel exposure; leaves open effects on platelet inhibition or outcomes in patients.
Does concomitant food intake affect the bioavailability of a single oral dose of clopidogrel in healthy male subjects?
Effect estimate: 6.1-fold increase in Cmax and 9.2-fold increase in AUC0-inf
Concomitant food intake, specifically a high-fat meal, substantially enhances the bioavailability of a single oral dose of clopidogrel in healthy subjects.
Nirogi et al. (2011) studied Healthy. Clopidogrel with food (standardized high fat breakfast) vs. Clopidogrel in fasted state was evaluated on Pharmacokinetic parameters (Cmax, AUC0-inf, t1/2) (6.1-fold increase in Cmax and 9.2-fold increase in AUC0-inf). Concomitant food intake substantially enhanced the bioavailability of a single oral dose of clopidogrel, increasing Cmax 6.1-fold and AUC0-inf 9.2-fold compared to the fasted state.
