Key result
PGI2 plus theophylline curbs shear-induced platelet aggregation but markedly increases high-shear lysis.
Why the study?
The effects of PGI2 combined with theophylline on human platelets subjected to shear stress and their clinical implications were not fully understood.

May inhibit shear-induced platelet aggregation in vitro; leaves open any clinical antithrombotic role.
Does pretreatment with PGI2 and theophylline affect shear-induced platelet aggregation and lysis in human platelet-rich plasma?
While PGI2 and theophylline reduce shear-induced platelet aggregation, they paradoxically increase platelet fragility and lysis at high shear stress, which may impact clinical applications.
Hardwick et al. (1981) studied Platelet response to shear stress. PGI2 and theophylline was evaluated on Shear-induced platelet aggregation, lysis, and serotonin release. Pretreatment of platelets with PGI2 and theophylline reduced shear-induced aggregation but caused a large increase in platelet lysis and serotonin release at stress levels ≥150 dynes/sq cm.
