Abstract Cellular communication relies on precise protein trafficking, yet conventional microscopy lacks the resolution to visualize nanoscale structures. STED microscopy combined with CRISPR-Cas9 endogenous tagging using SNAP- and Halo-tags enables high-resolution imaging of protein dynamics in their native environment, revealing detailed organization of secretory compartments, endosomes, and immune signaling organization.
