Vehicle-treated control recipients developed progressive proteinuria after 12 weeks, macrophage-dominated infiltration, dense ICAM-1 and cytokine expression (TNF-α, TGF-β, PDGF), and progressive interstitial fibrosis and vascular obliteration.
RS61443 treatment initiated at either day 0 or week 8 maintained normal graft function, virtually eliminated proteinuria, and markedly reduced morphological and immunohistological signs of chronic rejection.
Treated animals exhibited no major adverse effects, including leukopenia, anemia, thrombopenia, nephrotoxicity, or hepatotoxicity.
