Hirudin-loaded pectin-crosslinked porcine pericardium improves anti-calcification and hemocompatibility versus GA-fixed PP.
Why the study?
Does dialdehyde pectin-crosslinked and hirudin-loaded decellularized porcine pericardium improve matrix stability, anti-calcification, and biocompatibility compared to glutaraldehyde-fixed pericardium for bioprosthetic heart valves?
Does dialdehyde pectin-crosslinked and hirudin-loaded decellularized porcine pericardium improve matrix stability, anti-calcification, and biocompatibility compared to glutaraldehyde-fixed pericardium for bioprosthetic heart valves?
AP + Hirudin-PP is a promising, highly biocompatible and anti-calcific alternative to traditional glutaraldehyde-fixed porcine pericardium for bioprosthetic heart valves.
Should not yet change valve material selection; hypothesis-generating for pectin-hirudin pericardium as GA alternative.
results revealed that the AP + Hirudin-PP specimens recruited the minimum immune cells in the implantation experiment, and also presented an excellent anti-calcification effect. Overall, AP + Hirudin-PP was endowed with competitive collagen stability (compared with GA-fixed PP), excellent hemocompatibility, good HUVECs-cytocompatibility, low immunogenicity and outstanding anti-calcification, suggesting that AP + Hirudin-PP might be a promising alternative to GA-fixed PP and exhibited a bright prospect in the clinical applications of BHVs.

Hu et al. (2021) studied Bioprosthetic heart valves. Dialdehyde pectin-crosslinked and hirudin-loaded decellularized porcine pericardium (AP + Hirudin-PP) vs. Glutaraldehyde (GA)-fixed porcine pericardium (GA-fixed PP) was evaluated on Immune cell recruitment, anti-calcification effect, collagen stability, hemocompatibility, and cytocompatibility. Dialdehyde pectin-crosslinked and hirudin-loaded decellularized porcine pericardium demonstrated competitive collagen stability, excellent hemocompatibility, and outstanding anti-calcification compared to GA-fixed PP.
