Ribonuclease T1 generates circular RNA molecules by cleaving and ligating viroid-specific RNA transcripts.
Why the study?
The potential of RNase T1 to process viroid RNA transcripts by cleavage and ligation to form circular RNA molecules was not previously demonstrated.
RNase T1 can act as an RNA 'circulase' by cleaving and ligating viroid-specific RNA transcripts in vitro, suggesting potential roles in RNA processing and recombination.

RNase T1 circularizes viroid RNA in vitro; leaves open its role in RNA recombination pending in vivo studies.
Tsagris et al. (1991) studied this question. Ribonuclease T1 was evaluated on RNA cleavage and intramolecular ligation. Ribonuclease T1 processed a 406-nucleotide potato spindle tuber viroid-specific RNA transcript by cleaving out a 358-nucleotide product and ligating it into a circular RNA molecule.
