Key result
Endothelin 1 increases human myocyte contraction amplitude by ~90%.
Why the study?
The presence and functional role of endothelin receptors on cardiac myocytes, compared to vascular smooth muscle cells, was unclear.
Does endothelin 1 bind to and affect the contractility of human and rat cardiac myocytes?
Population
Intact rat myocardium and human ventricular samples from explanted hearts, isolated cardiac and smooth muscle cells from human and rat
Comparison
Endothelin 1 vs baseline physiological saline, phenylephrine (rat), and high extracellular calcium (>10 mM, human)
Design
Preclinical autoradiography and isolated cell functional contraction study

Endothelin-1 increases contractility in isolated human myocytes; extends rat data but leaves open in vivo relevance and therapeutic targeting.
Does endothelin 1 bind to and affect the contractility of human and rat cardiac myocytes?
Absolute Event Rate: 4% vs 2.1%
p-value: p=<0.001
Endothelin 1 receptors are present on human and rat cardiac myocytes and their activation significantly increases contraction amplitude.
Moody et al. (1990) studied Heart failure (explanted hearts for transplant). Endothelin 1 vs. Baseline (physiological saline) was evaluated on Contraction amplitude (% cell length) in human cardiac myocytes (p=<0.001). Endothelin 1 approximately doubled the contraction amplitude of isolated cardiac myocytes from 4.1% to 8.1% in rats (p<0.001) and from 2.1% to 4.0% in humans (p<0.001).
