Low-dose triple combination therapies reduce SBP up to ~20 mmHg vs placebo in mild-to-moderate hypertension.
Why the study?
The dose-responsiveness, efficacy, and safety of low-dose triple antihypertensive combination therapies in patients with mild-to-moderate hypertension had not been established.

Does low-dose triple antihypertensive combination therapy reduce systolic blood pressure in patients with mild-to-moderate hypertension?
Low-dose triple combination therapy (amlodipine, losartan, chlorthalidone) effectively reduces systolic blood pressure in mild-to-moderate hypertension with potentially fewer side effects like ankle edema compared to high-dose monotherapy.
Supports low-dose triple combinations for initial SBP management in mild-to-moderate hypertension; extends RCT evidence for combination regimens.

Hong et al. (2020) conducted an RCT in mild-to-moderate hypertension (n=248). low-dose triple combination therapies (amlodipine + losartan + chlorthalidone) vs. amlodipine 10mg, amlodipine 5mg, losartan 100mg, and placebo was evaluated on mean change in systolic blood pressure (SBP) from baseline to week 8. Low-dose triple combination therapies yielded placebo-corrected systolic blood pressure reductions of 14.9 to 19.5 mmHg in patients with mild-to-moderate hypertension.
