Captopril's dual actions in rat arteries should not yet inform clinical dosing; leaves open whether angiotensin-independent effects occur at therapeutic concentrations in humans.
Does captopril attenuate adrenergic vasoconstriction in isolated rat mesenteric arteries?
Captopril attenuates sympathetic vasoconstriction through both angiotensin-dependent mechanisms at low concentrations and angiotensin-independent mechanisms at high concentrations.
Collis et al. (1981) studied this question. Captopril was evaluated on Vasoconstrictor response evoked by sympathetic nerve stimulation. In isolated rat mesenteric arteries, captopril antagonized the potentiating effect of angiotensin I on sympathetic nerve stimulation and, at high concentrations, directly antagonized responses to noradrenaline.
