Key result
rHuIFN-alpha 1 shows markedly lower human antiviral activity versus other interferons despite unique murine efficacy.
Why the study?
The study aimed to elucidate the nature and relative antiviral activity of different recombinant human interferon subtypes across various cell cultures.
Population
Bovine cells, human fibroblasts, human WISH cells, murine L 929 cells infected with VSV
Comparison
rHuIFN-alpha1 vs rHuIFN-alpha2 vs human leukocyte interferon
Design
Preclinical in vitro microtitre assay measuring antiviral activity

Requires caution extrapolating murine interferon activity to human cells; leaves open standardized human-cell assays for reliable evaluation.
The antiviral activity of recombinant human interferon subtypes varies significantly depending on the cell line used for the assay.
Tonew et al. (1989) studied this question. rHuIFN-alpha 1 vs. rHuIFN-alpha 2 and human leukocyte interferon was evaluated on 50% inhibition of the cytopathic effect of vesicular stomatitis virus (VSV). rHuIFN-alpha 1 had 45 to 212 times lower antiviral activity on human cells compared to other interferons, but was the only active interferon on murine L 929 cells.
