Key result
Adding nifedipine to atenolol slightly increases platelet dense granule release without affecting coagulation.
Why the study?
The effects of atenolol alone and in combination with nifedipine on platelet behaviour and blood coagulation in healthy volunteers were unclear.
Does treatment with atenolol or a combination of atenolol and nifedipine affect platelet behaviour and blood coagulation in healthy volunteers?
Population
Healthy volunteers
Comparison
Atenolol vs placebo; atenolol vs atenolol and nifedipine combination
Design
Two double-blind crossover studies

May increase ex vivo platelet reactivity with combination therapy; hypothesis-generating for clinical thrombotic risk and needs larger trials.

Does treatment with atenolol or a combination of atenolol and nifedipine affect platelet behaviour and blood coagulation in healthy volunteers?
Combined treatment with atenolol and nifedipine slightly increases platelet dense granule release ex vivo in healthy volunteers, without affecting coagulation parameters.
Horn et al. (1992) conducted an RCT in Healthy volunteers. Atenolol and fixed combination of atenolol and nifedipine vs. Placebo and atenolol alone was evaluated on Platelet behaviour and blood coagulation. Combined treatment with atenolol and nifedipine caused a small but significant increase in platelet dense granule release ex vivo compared to atenolol alone, with no effect on coagulation.
