Measured systolic blood pressure and left ventricular mass index (LVMI).
Quantified MMP-2 and MMP-9 activity via gelatin zymography, and measured MMP-2, MMP-9, TIMP-1, and TIMP-2 mRNA and protein levels via RT-PCR and Western blot.
Amlodipine monotherapy and combination therapy significantly lowered blood pressure and left ventricular mass index in SHR rats compared to untreated controls (P < 0.05).
Elevated myocardial MMP-2 and MMP-9 mRNA, protein expression, and enzymatic activity in SHR rats were significantly reduced by amlodipine, atorvastatin, and most potently by combination therapy (P < 0.05).
Elevated myocardial TIMP-1 and baseline TIMP-2 levels in SHR rats were not significantly altered by amlodipine, atorvastatin, or combination treatment.
