Structured PICO

Does sildenafil pretreatment improve postresuscitation myocardial dysfunction and energy metabolism in a porcine model of cardiac arrest?

P
Population
24 inbred male Wuzhishan (WZS) miniature piglets ages 11 to 13 months, average weight 30 ± 2 kg, subjected to 8 minutes of untreated ventricular fibrillation followed by 15 minutes of open-chest cardiopulmonary resuscitation.
I
Intervention
Sildenafil 0.5 mg/kg administered once intraperitoneally 30 minutes prior to ventricular fibrillation (VF).
C
Comparator
Saline (0.9% NaCl) administered in the same manner and volume, and a sham operation group.
O
Outcome
Severity of postresuscitation myocardial dysfunction and energy metabolism (myocardial interstitial fluid concentrations of glucose, lactate, pyruvate, glutamate, and glycerol) up to 24 hours after resuscitation.surrogate

In a porcine model of cardiac arrest, pretreatment with sildenafil improved 24-hour survival, hemodynamics, and myocardial energy metabolism, suggesting a potential cardioprotective role against ischemia/reperfusion injury.
