Oral calpain inhibitor BDA-410 attenuates Angiotensin II-induced aortic aneurysm formation in mice.
Why the study?
Does calpain inhibition with BDA-410 prevent Angiotensin II-induced ascending and abdominal aortic aneurysms in calpain-1 deficient and wild-type mice?
Does calpain inhibition with BDA-410 prevent Angiotensin II-induced ascending and abdominal aortic aneurysms in calpain-1 deficient and wild-type mice?
p-value: p=<0.05
Calpain-2 compensates for the loss of calpain-1 in AngII-induced aortic aneurysms, and pharmacological inhibition of both isoforms attenuates aneurysm formation in mice.

Calpain inhibition attenuates AngII-induced ascending aortic aneurysms in mice irrespective of calpain-1 status; leaves open isoform-specific targeting as a therapeutic strategy in humans.
Subramanian et al. (2013) studied Angiotensin II-induced ascending and abdominal aortic aneurysms. BDA-410 (calpain inhibitor) vs. Vehicle was evaluated on Angiotensin II-induced ascending and abdominal aortic aneurysm formation (p=<0.05). Oral administration of the calpain inhibitor BDA-410 significantly attenuated Angiotensin II-induced ascending and abdominal aortic aneurysm formation in both calpain-1 +/+ and -/- mice.
