This novel method allows for the selective loading of indo-1 free acid into the cytosol of cardiac myocytes to accurately assess intracellular calcium concentrations.

A novel method to load indo-1 free acid into rat ventricular cells yielded weighted mean cytosolic Ca2+ concentrations of 138 nM in quiescent cells and 435 nM at the transient peak.
