Key result
Open-label aliskiren/amlodipine achieves BP control in ~74% of patients, with frequent peripheral edema.
Why the study?
Most patients with hypertension require antihypertensive combination therapy to achieve BP control, prompting investigation of the direct renin inhibitor aliskiren combined with the calcium channel blocker amlodipine.
Does aliskiren/amlodipine combination therapy safely reduce blood pressure in patients with hypertension?

Does aliskiren/amlodipine combination therapy safely reduce blood pressure in patients with hypertension?
Long-term combination therapy with aliskiren and amlodipine is well tolerated and effectively lowers blood pressure in hypertensive patients.
Littlejohn et al. (2009) studied hypertension (n=556). aliskiren and amlodipine was evaluated on long-term safety and tolerability, and BP-lowering efficacy. Open-label aliskiren/amlodipine 300/10 mg for 54 weeks reduced mean BP by 24.2/15.5 mmHg (74.3% achieved BP control), with peripheral edema occurring in 22.7% of patients.
