Following the first oral dose, sitting diastolic blood pressure decreased significantly with Ro 42-5892 versus placebo at peak (−10.2 ± 1.2 mm Hg vs −0.4 ± 2.0 mm Hg, P < .01) and trough (−6.9 ± 1.8 mm Hg vs 1.7 ± 0.9 mm Hg, P < .01).
Trough diastolic blood pressure reductions remained significant after the 7th dose (−5.1 ± 1.6 mm Hg vs −0.2 ± 1.0 mm Hg, P < .05) and 8th dose (−5.4 ± 1.3 mm Hg vs 2.3 ± 1.2 mm Hg, P < .01).
Oral and intravenous Ro 42-5892 induced prolonged suppression of plasma renin activity and a transient increase in immunoreactive renin levels.
